Linsitinib

Linsitinib is an oral, ATP-competitive dual IGF-1R and insulin receptor (InsR) inhibitor, blocking ligand-stimulated receptor autophosphorylation and downstream IRS-1/PI3K/AKT signalling. IGF-1R co-inhibition with InsR prevents the compensatory hyperinsulinaemia-driven InsR activation that limits selective IGF-1R inhibitors. Active in Ewing sarcoma and adrenocortical carcinoma models.
